Route 16 is a secondary leg of the Trans-Canada Highway (TCH). The 51 kilometre-long route goes from an interchange with the mainline TCH (Route 2) near the Nova Scotia border in Aulac, to the Confederation Bridge at Cape Jourimain, connecting New Brunswick to Prince Edward Island. Port Elgin is the only major community along the route.

Route 16 begins near the terminus of Route 2 at the Nova Scotia border at a cloverleaf interchange, traveling east. The highway passes through several small communities located along the border, as it traverses rural Westmorland County. The first major intersection is a y intersection with Main Street just west of Baie Verte, which intersects Route 970 in the town. The highway travels north of the town, paralleling Route 970 until a roundabout interchange with Route 970 and the eastern terminus of Route 15 in Port Elgin. Route 16 continues northeast, intersecting Route 960 as it continues east to the town of Bayfield, where the highway meets Route 955. The final interchange on Route 16 provides access to the Cape Jourimain Nature Centre, before the highway continues east along the Confederation Bridge into Prince Edward Island and Route 1.[1]
Route 16 has been part of the New Brunswick highway network since the 1920s. The only major changes to the route have been a Port Elgin bypass and approaches to the Confederation Bridge.
